ABSTRACT

Chronic intestinal pseudo-obstruction (CIPO) is caused by a plethora of primary and secondary causes, dominantly involving the neuromuscular tissue, interstitial cells of Cajal, or the connective tissue framework. The lack of the connective tissue framework, known as desmosis, is evaluated by Masson's trichrome (MT) or picrosirius red stains, both of which are recommended in the London classification. We evaluated the orcein stain in detecting desmosis in comparison to the MT stain. We performed both orcein and MT stains in six previously published cases of complete or partial desmosis along with six age-matched controls. Our results showed comparable results of the orcein stain as compared to the MT stain. Additional advantages of lower cost and a clearer background in orcein stain were noteworthy, whereas MT stain can be used for the detection of additional pathology. We believe that orcein stain can be used as a cheap alternative in resource-limited settings.

ABSTRACT

@#Intravascular Papillary Endothelial Hyperplasia (IPEH) is a rare benign vascular lesion of the skin and subcutaneous tissues that results from the proliferation of endothelial cells within a blood vessel. In this article, we present the case of an IPEH mimicking a Sister Mary Joseph Nodule on the umbilicus of a 53-year old female patient with ovarian cancer. A diagnostic workup was performed with a computerized tomography of abdominal cavity and pelvis showing an expansive cystic tumor formation with probable ovarian origin. Two separate histopathologic readings were done on the cutaneous lesion which revealed contradicting findings of benign versus malignant tumors. Immunohistochemical stains done showed that the lesion was positive for ERG and negative for epithelial differentiation markers. No surgical intervention was done at the time of consultation as a cutaneous metastasis was primarily considered initially. It is crucial to rule out a diagnosis of Sister Mary Joseph nodule, especially in a background of ovarian carcinoma as it may mimic vascular lesions occurring on the umbilicus. Immunohistochemical staining is a significant tool to precisely diagnose such lesions so that it is neither inadequately nor aggressively managed.

ABSTRACT

SUMMARY: Peri-implantitis is an inflammatory lesion of bacterial etiology characterized by inflammation of the mucosa and bone loss. Chronic inflammation is characterized by neovascularization and collagen neoformation. Mast cells have been shown to participate in the inflammatory process by releasing mediators and proteases such as chymase and tryptase, important in the collagen neoformation process. Although a higher percentage of collagen has been described in periodontal disease, the literature is scarce about the percentage of collagen in peri-implantitis. The aim of this study was to quantify the percentage of collagen fibers present in the peri- implant soft tissue of patients with peri-implantitis lesions. A descriptive observational cross-sectional study was performed. Samples of peri-implant soft tissue were collected from eleven patients with peri-implantitis and then processed by Masson's Trichrome Technique. In microscopic analysis, collagen fibers were observed in all samples, with an average percentage of 39.89 %, standard deviation of 17.02 %, with a minimum value of 8.99 % and a maximum value of 75.65 % density. From these results, it can be concluded that in human peri-implantitis lesions with bone loss greater than 50 %, there is an important percentage of collagen fibers, which is interpreted as connective tissue in a permanent process of reparative response, in the presence of inflammatory infiltrate.

ABSTRACT

BACKGROUND: In current studies regarding skeletal muscle blunt contusion model, rats are often taken as the experimental object. The authors believe that rabbits are the better experimental object to observe the macroscopic and imaging changes after blunt contusion. OBJECTIVE: Based on the self-made heavy-duty smashing instrument as the experimental basis, to replicate the skeletal muscle injury model according to different strike heights, which is expected to provide a reference basis for the subsequent replication of the rabbit gastrocnemius blunt contusion model. METHODS: Thirty-three New Zealand rabbits were randomly divided into four groups: normal group (n=3), 75 cm strike group (n=10), 50 cm strike group (n=10), 25 cm strike group (n=10). Except for the normal group, all strikes were performed with different strengths. The severity of skeletal muscle damage after different strikes was compared through gross observation, hematoxylin-eosin staining, Masson staining, and ultrasound imaging. RESULTS AND CONCLUSION: The 50 cm height blow can cause moderate damage to the skeletal muscle, and the natural recovery time is over 21 days, which is suitable for the establishment of skeletal muscle blunt contusion model. The 75 cm height blow is likely to cause fractures, resulting in more deaths. The 25 cm height blow can cause mild damage that can be recovered within 7 days, which is not suitable for the establishment of skeletal muscle injury model.

ABSTRACT

Background: Oral submucous fibrosis (OSMF) is graded according to various histological factors which include the epithelial changes and the connective tissue changes. These features could be identified in routine Hematoxylin and Eosin (H and E) staining in shades of pink. However, the use of special stains provides a contrast to various connective tissue components thereby aiding in better visualization of these connective tissue changes in advanced OSMF cases. Objective:To compare and evaluate muscle involvement and degeneration in advanced oral submucous fibrosis using three different stains namely, H&E, Van Gieson, and Masson's Trichrome. Material and Methods: 10 Formalin-fixed paraffin-embedded tissue sections of advanced OSMFwere stained using 3 different stains namely Hematoxylin and Eosin (H&E), Van-Gieson, and Masson trichrome. The results obtained were tabulated and statistically analyzed using Kruskal Wallis ANOVA. Results: The hyalinization and fibrosis involving the skeletal muscle were better visualized in Masson's Trichrome but were not statistically significant. The muscle degeneration in deeper areas was better visualized in Masson's trichrome when compared to the H&E and Van Gieson. Conclusion: Hematoxylin and eosin stains all the connective tissue components in various shades of pink, use of special stains bestows contrast between different components of connective tissue, thus improvising grading of OSMF. Masson's trichrome stain can be used as a single adjunct to hematoxylin and eosin stain as changes in the superficial and deeper connective tissue could be ascertained (AU)

ABSTRACT

Objective@#To investigate the effect of low level laser on osteoclast and collagen fiber remodeling during the process of tooth retention after tooth movement in rats and to provide the experimental basis for clinical application.@*Methods @# In total, 20 eight-week-old Wistar rats were selected to establish a mesial movement model of the maxillary first molar and then randomly divided into four groups after the appliance was removed. In total, 5 rats were included in each group, including baseline group (without force as blank control), control group (without any intervention after removing the force appliance), retention group (teeth were wrapped with orthodontic ligature wires that were screwed into hemp flower as fixed retention to maintain the space between the first molar and incisor after appliances were removed) and retention and low energy laser irradiation group (teeth were wrapped with the orthodontic ligature wires that were screwed into hemp flower as fixed retention and low energy laser irradiation was applied on days 0, 3, 6, 9 and 12 after appliance removal). Two weeks later, all the rats were sacrificed and the first molar tissue blocks of each group were collected. The distribution of osteoclasts and collagen fiber were studied by HE staining, TRAP staining and Masson staining to illustrate the process of alveolar bone and collagen fiber remodeling.@*Results @# Two weeks after appliances were removed, collagen fibers were deposited on both sides of the root in the baseline group, but no osteoclasts were observed in the distal side of the root. In the control group, collagen fibers on the two sides of the root were not obvious and osteoclasts were active on the distal side. In the retention group, collagen fibers were obvious on the two sides of the root and the osteoclasts on the distal side were less active than the control group. Regarding the retention and low energy laser irradiation group, collagen fibers were significantly obvious and osteoclasts were not seen. The difference was statistically significant between the retention and low energy laser irradiation group and the other three groups (P<0.05). @*Conclusion@#These results suggest that fixed retention with simultaneous low level laser can effectively promote the synthesis of collagen fibers and inhibit the activity of osteoclasts during the process of tooth retention after movement, thus reducing the possibility of molar recurrence.

ABSTRACT

SUMMARY: Reproductive dysfunction is a complication for many diseases and toxins. Its early diagnosis and treatment are immensely important. Here the morphological histoarchitecture changes in early testicular and cauda toxicity before and after treatment with angiotensin receptor blockers were evaluated. Low-grade testicular damage was induced using thioacetamide (TAA, 50 mg/kg/day) intraperitoneally for two weeks in rats. The rats were randomly divided into four groups (n = 8) treated daily orally for three weeks as follows: Normal control (distilled water), TAA (positive control), TAA+candesartan (0.2 mg/kg) and TAA+losartan (7.5 mg/kg). Serum testosterone and testicular malondialdehyde and glutathione were measured. The changes in histoarchitecture of testis and cauda epididymis were evaluated by hematoxylin and eosin for general structure, Masson's trichrome for collagen, periodic acid Schiff for basement membrane, and caspase-3 and proliferating cell nuclear antigen (PCNA) for immunohistochemical analysis. The TAA-rats showed decreases of serum testosterone and testicular glutathione, increases in testicular malondialdehyde, degenerative changes and apoptosis in germ cells, thickening of tubular basal lamina and increases in expression of caspase 3, and decreases in expression of PCNA. The ARBs (candesartan and losartan) significantly reversed these changes with non-significant differences in-between. Treatment with ARBs (candesartan and losartan) significantly reversed TAA-induced low-grade testicular and cauda toxicity in rats. This could be potentially useful for early treatment of male patients with occupational toxicant-induced reproductive dysfunction especially if they are using ARBs for other comorbidities.

ABSTRACT

Background: Identification and differentiation of stromal hard tissue components is a challenging task. Numerous methods of demonstrating these components have been worked upon in the past. Although some of the methods have been successful, there are many drawbacks of employing them. The need of the hour, therefore is to develop and use a simple, rapid and cost-effective method of identifying stromal hard tissues as they may signify an important change in the diagnosis of the pathology. Our aim is therefore to observe the usability of tetrachromic VOF stain over Hematoxylin and Eosin and Masson's Trichrome in routinely encountered head and neck pathologies. Materials and Method: Routine cases such as Central and peripheral ossifying fibromas, osteomas, giant cell granulomas, osteomyelitis and malignancies like osteosarcomas were retrieved from the department archives and 3 sections from each block were prepared to stain with H and E, Masson's trichrome and modified tetrachromic VOF stains respectively using standard staining protocol. Results: Tetrachromic VOF takes an upper hand in stromal hard tissue differentiation irrespective of the pathology. Conclusion: Modified tetrachromic VOF is simple, cost-effective method and can be employed for diagnosis of cases with hard tissue differentiation within the stroma on routine basis.

ABSTRACT

Objective: To explore a rapid histological preparation method to observe morphology and composition distribution of tendon collagen fascicle and endotendinum. Methods: Taking porcine superflexor tendon of foot as an example, tendons were sliced into sections with 6 μm by frozen section technology, after which general observation of the section integrity was carried out. After fixed with 10% neutral buffered formalin and performed with HE staining, the tissue integrity and ice crystal formation were observed under microscope. Sections were then divided into 5 groups by different methods of dyeing. Group A: Priodic acid-Shiff (PAS) staining; group B: Masson staining; group C: reticular fibers staining; group D: immunohistochemical and immunofluorescent staining of type Ⅲ collagen; group E: the sections were baked at 65℃ for 10 minutes and stained with Masson. The composition distribution of tendon collagen fascicle and endotendinum in different groups were observed. Results: From general observation, the frozen section of tendon tissue was complete and continuous. Although the tissue integrity in the tendon sections could be seen and no ice crystal was formed, the composition distribution could not be identified by HE staining. The entire tendons in groups A, B, and C were dyed, and the composition distribution of collagen fascicle and endotendinum could not be identified. The endotendinum in group D was stained weakly positive for type Ⅲ collagen alone, and the two components were differentiated dyed but the contrast was not obvious. In group E, the collagen fascicle and endotendinium were differentiated dyed and the two components in tendon tissue were clearly visible. Conclusion: The morphology and the composition distribution of tendon collagen fascicle and endotendinum can be characterized rapidly and accurately, using a combination of baking at 65℃ for 10 minutes and Masson staining after porcine superflexor tendons were sliced by frozen section technology.

ABSTRACT

Intravascular papillary endothelial hyperplasia or Masson's hemangioma is a rare vascular tumor. The reactive proliferation of endothelial cells in this disease mimics other benign or malignant vascular proliferation such as angiosarcoma or Kaposi's sarcoma. It is important to make an accurate distinction to avoid confusion with these malignant tumors. This would facilitate a proper diagnosis, which is essential so that the patient is not subjected to unnecessarily aggressive or inappropriate treatment.

ABSTRACT

Abstract Background: Fibrosis is present in several pathologies associated with mammary carcinogenesis. Objective: To evaluate and quantify the fibrosis present in malignant and benign mammary neoplasms in bitches. Methods: Eighty-three samples were divided according to histopathological diagnosis into benign (n= 21) and malignant (n= 62) neoplasms. Haematoxylin-eosin and Masson's trichrome were used to locate the connective tissue, and the extent of fibrosis was assessed with image software. Results: Benign neoplasms were classified into adenomas (cystic, complex, and tubular), benign mixed tumor, and ductal and lobular hyperplasia. Malignant neoplasms were classified as carcinomas (complex, mixed tumor, in situ tubular, tubulopapillary, and solid). Grade I was the most prevalent histopathological class, followed by grade II and III. Fibrosis was classified as severe, moderate, or discrete. No significant (p >0.05) difference was observed for the percentage of fibrosis between malignant and benign group neoplasms. However, difference (p=0.028) was found for fibrosis percentage between histopathological subtypes of tumors. The benign subtype of lobular hyperplasia presented differences between cystic adenoma and benign mixed tumor. The in situ malignant tubular carcinoma subtype presented differences between solid and tubulopapillary carcinoma. Conclusions: Fibrosis in canine mammary tumors can be estimated with Massons's trichrome staining.

ABSTRACT

Intravascular papillary endothelial hyperplasia (IPEH) is a rare cause of orbital mass which can affect healthy individuals of any age/sex/race/region. It usually requires a histopathological examination, characterized by the appearance of papillary proliferation of vascular endothelial cells within the lumen of the blood vessel, for the final diagnosis. One should be aware that this is a highly vascular lesion which can cause excessive intraoperative bleeding and incomplete removal can lead to recurrences. IPEH of the orbit/eyelid has been reported in few parts of the world as isolated case reports but none from India so far.

ABSTRACT

Objective; To investigate the dynamic pathological characteristics of kidney in the db / db mice with type 2 diabetes mellitus and to clarify the pathogenesis and development mechanism of diabetic nephropathy, and to provide the experimental evidence for the further study on diabetic nephropathy. Methods: The male SPF db/db mice aged 7 - 8 weeks were selected as model group (n=16) and the db/m mice with the same ages were selected as control group (n=16). The body weights and levels of fasting blood glucose (FBG) of the mice were detected after 8, 16, and 32 weeks. Then eight mice in each group were sacrificed and the kidney tissue was dissected at 8, 16 and 32 weeks. HE and Masson staining were used to observe the pathomorphology of the kidney tissue; the ultrastructures of kidney tissue were observed under electron microscope. Results: Compared with normal group, the body weights of the mice in model group at 8, 16 and 32 weeks were significantly increased (P<0. 01), the FBG levels were significantly increased (P<0. 01), and the double kidney indexes were significantly decreased (P<0. 01); the double kidney index of the mice in model group at 32 weeks was lower than that at 16 weeks (P< 0. 05). The HE and Masson staining results showed that the morphological changes of kidney tissue of the mice at 16 weeks were significant, the glomerular volume was expansion and the kidney tubular epithelial cells were edema, which turned serious at 32 weeks. A large number of blue dye substances in the kidney tissue was found. Under electron microscope, the glomerular basement membranes got thickened, the foot processes got fused and the number of mitochondria in kidney tubular epithelial cells was reduced and swell was found in the mice in model group at 16 weeks; the lesions in the kidney tissue were serious at 32 weeks, and the collagenous fiber was visible at 32 weeks. Conclusion: The pathological changes in the kidney tissue of the db/db mice at 16 weeks are significant, such as glomerular basement thickening and footwork fusion. The kidney tissue of the db/db mice at 32 weeks shows prominent fibrosis.

ABSTRACT

@#Introduction: This study provides a description of development of various components of the human ureter at different gestational ages. Methods:Measurements of the length of the ureter from pelvi-ureteric junction to vesico-ureteric junction, length of its intravesicular portion and angle made by the ureter at the entry into the bladder with respect to the internal ureteric orifice were taken. Transverse sections of the ureter were taken at the upper end, close to the pelvi-ureteric junction, and at the midpoint between the pelvis of the kidney and the vesico-ureteric junction. These were subjected for processing and stained with Haematoxylin Eosin and Masson’s Trichrome. They were then studied under the light microscope. Results: The length of the ureter from the pelvi-ureteric junction to the vesico-ureteric junction showed a highly positive correlation with the gestational age (GA). The angle of entry of the ureter to the bladder decreased as the GA increased. The microstructure of the ureter in both the sections showed development of transitional epithelium with an increase in layers by 33 weeks. At 18 weeks, the smooth muscle layer was in a single layer with an abundance of interspersed collagen fibers. By 33 weeks, the thickness of the wall increased significantly with a decrease in collagen. There was no trace of longitudinal muscle fibers even upto 33 weeks. Conclusion: Epithelial, muscular and connective tissue components displayed significant changes during intra uterine development.

ABSTRACT

It was fi rst described by Massonin 1923 as intravascular vegetating hemangio endothelioma, andlater received diff erent names, like «intravascular angiomatosis¼,«Masson’s pseudoangiosarcoma¼ and «intravascular papillaryendothelial hyperplasia¼; fi ndings in the mouth are rare, and when present, they are generally in the lip. The clinical case presented is a78-year-old patient with a bilobular asymptomatic lesion in gingivawith a slow and progressive 10-year evolution. A presumptive clinicaldiagnosis of peripheral giant-cell granuloma was issued, followed byan excisional biopsy and histopathology study to confi rm the diagnosis;it had been correctly diagnosed as IPEH, which was confi rmed byimmuno histochemistry. This lesion can be easily confused with vascular malignancies (angiosarcoma, Kaposi sarcoma) both clinically andhistologically. The intention of this work is, through a clinical case,to describe the importance of a correct diagnosis by a proper clinical examination, detailed microscopic observation and interdisciplinary communication between the surgeon and the pathologist, in order toavoid a misdiagnosis of this benign entity with other pathologies tha trequire more aggressive and invasive treatments, inadequate for IPEH.

ABSTRACT

Background: Oral submucous fibrosis (OSMF), a potentially malignant oral disorder has the highest rate of malignant transformation of about 7-13%. The connective tissue changes that occur in this disease are characteristic and are stained with special stains. Objective: The study was done to compare common and special stains under light microscopy and polarizing microscopy to evaluate the levels of fibrosis in oral submucous fibrosis and assess the type of collagen present in the stromal area. Materials and Methods: Fifty tissue blocks were selected from the archives and were prepared and stained with H&E, Masson's trichrome, Van Gieson and Picrosirius red and studied under light microscope and polarizing microscope respectively. Results: H and E stained slides were useful in diagnosing the lesion but was not able to highlight the level of fibrosis. Masson's trichrome and Van Gieson stained slides showed the depth of the lesion which extended even to the deeper muscle layer. The type of collagen present was definitively seen by the birefringence in polarizing microscopic study. Interobserver variation was less and all the values regarding the effectiveness of the special stains in detecting the level of fibrosis were statistically significant. Conclusion: Special stains can be used routinely in laboratories to demonstrate connective tissue lesions especially in cases of OSMF. Depth of the lesion and the area of involvement help in treatment planning to be delivered. Large scale studies with more categories and inclusion criteria are required along with the special stains to assess the other alterations in OSMF

ABSTRACT

Dyke-Davidoff-Masson syndrome ( DDMS) is a rare epilepsy syndrome which is characterized by cerebral hemiatrophy with ipsilateral compensatory skull changes and contralateral hemiplegia recurrent .Here we reported a case of which the curative effect turned out to be unsatisfied after conservative treatment , physical examination re-vealed mental retardation , facial asymmetry , mild right hemiparesis , MRI scan of the head showed left cerebral hemiatrophy , calvaria thickening , PET-CT showed less functional left cerebral hemisphere .The patient finally un-derwent multi-lobe disconnection to relieve recurrent seizure , whose seizure was well controlled in the following up of one year .

ABSTRACT

Objective To compare hematoxylin and eosin(HE) staining,Masson's staining and picric sirius red(PSR) staining in evaluation of cardiac fibrosis. Methods Twenty adult sprague dawley (SD) rats were given subcutaneous injection of isoprenaline for 5mg/(kg · d) for 3 weeks, which induced cardiac fibrosis model successfully. In order to observing the fibrotic areas and collagen deposition,we prepared paraffin sections by using the heart specimen of rat,and carried out HE staining,Masson staining and PSR staining respectivelly for the special staining. Results All staining methods could clearly show the collagen fibers and fibrosis stage. We could clearly distinguish the type Ⅰ and type Ⅲ collagen fibers through PSR staining with immunoflurescent techniques, but not Masson and HE staining. Conclusion The results demonstrated that PSR staining is better than HE and Masson staining to be used for the evaluation of degree and type of proliferation of collagen fibers. It has important signification for treatment and prevention of cardiac fibrosis.
